{"id":967,"date":"2014-06-18T12:28:12","date_gmt":"2014-06-18T19:28:12","guid":{"rendered":"http:\/\/sop.washington.edu\/?page_id=967"},"modified":"2025-04-22T09:30:50","modified_gmt":"2025-04-22T16:30:50","slug":"instrumentation","status":"publish","type":"page","link":"https:\/\/staff.washington.edu\/twentzel\/wordpress\/department-of-pharmaceutics\/research\/pharmacokinetics-and-modeling-lab-pk-lab\/instrumentation\/","title":{"rendered":"Instruments"},"content":{"rendered":"<p class=\"intro\">The School of Pharmacy Bioanalysis and Pharmacokinetics Lab is equipped with LC-MS\/MS and LC-UV systems available for use by students, staff, faculty, and outside users.<\/p>\n<h2>Triple Quadrupole Instruments<\/h2>\n<p><strong>Agilent 1290 UPLC + Agilent 6460<\/strong><\/p>\n<p>Use-cases: Small molecule drugs and drug metabolites<\/p>\n<p>An Agilent 1290 series HPLC coupled to an Agilent 6460 triple-quadrupole mass spectrometer. It is used for quantitation of xenobiotics and their metabolites in the concentration range of high fg\/ml to low pg\/ml range. The mass spectrometer is equipped with an electrospray ionization source and can handle both negative and positive ions. The 1290 HPLC allows the use of HPLC and UHPLC columns for high-speed chromatography. Standard sample vials and 96-well plate formats are compatible.<\/p>\n<p><strong>Waters Xevo TQ Absolute<\/strong><\/p>\n<p>Use-cases: Peptide quantification, Small-molecule drugs and drug metabolites<\/p>\n<p>The Waters Xevo TQ Absolute offers the highest sensitivity and specificity of the HPLC systems in the lab. The instrument is equipped with Waters\u2019 IntelliStart software, which can optimize settings on the mass spectrometer for highly sensitive analysis. The Waters Xevo system has two liquid chromatography options:<\/p>\n<p><strong>Waters ACQUITY UPLC M-Class Chromatography System<\/strong><\/p>\n<p>The ACQUITY UPLC M-Class System is the lab\u2019s nano- to microflow system. The M-Class is capable of flow rates from 200 nL\/min to 100 \u00b5L\/min and a range of column inner diameter (I.D.) from 75 \u00b5m to 1.0 mm. Use of the M-Class comes couples with a nanoflow-compatible source on the Xevo mass spectrometer. Standard sample vials and 96-well plate formats are compatible.<\/p>\n<p><strong>Waters ACQUITY UPLC I-Class System<\/strong><\/p>\n<p>The ACQUITY UPLC I-Class System is capable of separations at flow rates ranging from 0.1 to 2 mL\/min. The I-Class allows the use of HPLC and UHPLC columns for high-speed chromatography. Standard sample vials and 96-well plate formats are compatible.<\/p>\n<h2>HPLC-UV Instruments<\/h2>\n<p><strong>Agilent 1290 UPLC + Diode Array Detector<\/strong><\/p>\n<p>Use-cases: Chromophore-containing small molecules<\/p>\n<p>An Agilent 1290 UPLC equipped with a diode array UV lamp capable of measuring wavelengths between 100 to 400 nm. The detector is capable of monitoring and recording up to eight wavelengths simultaneously while capturing full spectrum data. The 1290 HPLC allows the use of HPLC and UHPLC columns for high-speed chromatography.
