Phosphate regulates cementum (cem) formation. I.Development: control of PPi/Pi locally is critical for development of a functional periodontium. Absence of Pi (TNAP null, hypophosphatasia) results in minimal cementum formation with exfoliation of teeth, in humans. In contrast, lack of PPi appears to favor cementum formation, e.g. ank mutant mice, with normal development of the PDL and bone. II. Regeneration: We predict that controlling the ratio of PPi/Pi locally in favor of Pi during regenerative procedures, will cementum formation and, subsequently, regeneration of the periodontium.
